Abstract

Background: Acute liver injury (ALI) is a rapid deterioration of liver function over a period of days to weeks in individuals without pre-existing liver disease , it results from damage to hepatocytes leading to inflammation ,hepatocellular death through necrosis or apoptosis and impaired liver function.
Objective:Cisplatin is a widely used chemotherapeutic agent for the treatment of various carcinomas and sarcomas, However, its clinical use is limited by several adverse effects, including hepatotoxicity .Cisplatin-induced liver injury occurs mainly through oxidative stress characterized by excessive production of reactive oxygen species (ROS) and depletion of endogenous antioxidant defenses, these lead to destruction of hepatic tissue architecture, inflammation, and hepatocyte death . Cilnidipine and Nicardipine are dihydropyridine calcium channel blockers with potential antioxidant and anti-inflammatory properties. Therefore, the present study aimed to compare the hepatoprotective effects of this drugs against cisplatin-induced liver injury in rats.
Methods: Thirty six male albino rats were divided into six group, with six rats in each group groupI (negative control) received corn oil orally (1ml/kg) for 7 consecutive days ;group II (positive control) received corn oil orally (1ml/kg) for 7 consecutive days and on day 5 injected with a single IP injection of 7mg/kg Cisplatin ;groupIII received cilnidipine orally (10mg/kg daily for 7 consecutive days) ;groupIV received nicardipine orally (24mg/kg daily for 7 consecutive days) ;groupV received cilnidipine orally (10mg/kg for 7 consecutive days), and on day 5, rats injected with a single intraperitoneal injection of cisplatin (7 mg/kg) ;group VI received nicardipine orally (24mg/kg for 7 consecutive days), and on day 5,rats injected with a single intraperitoneal injection of cisplatin (7 mg/kg)
Results: In groupV and groupVI, the administration of Cilnidipine and Nicardipine both significantly at the same effect level decreased serum liver enzymes (ALT, AST, and ALP) and inflammatory cytokines (TNF-α and IL-1β), while increasing antioxidant enzyme activities (SOD and catalase) compared with the positive control group.
Conclusion: The results showed that both drugs offered significant protection against hepatic injury induced by cisplatin at the same effect levels.
