Abstract
Abstract
Background: Methotrexate (MTX) is an effective antineoplastic agent, but its clinical use may be limited by adverse effects, including nephrotoxicity and intestinal mucositis. Oxidative stress, inflammation, and apoptosis contribute to MTX-induced tissue injury. Biochanin A, a naturally occurring isoflavone, possesses antioxidant and anti-inflammatory properties and may provide tissue protection.
Objective: To investigate the protective effects of two doses of Biochanin A compared with vitamin E against MTX-induced nephrotoxicity and jejunal mucositis in male Wistar rats.
Methods: Forty-nine male Wistar rats will be randomly allocated into seven groups (n=7). MTX (7 mg/kg, intraperitoneally) will be administered on days 3–5. Biochanin A (10 or 20 mg/kg) or vitamin E (100 mg/kg) will be administered orally for five days. Kidney and jejunal injury will be assessed using serum BUN and creatinine, oxidative stress markers (MDA and GSH), inflammatory and anti-inflammatory markers, MPO, Nrf2/HO-1 and NF-κB gene expression, apoptotic markers (cleaved caspase-3 and BCL-2), mucin levels, diarrhea score, body weight, and histopathology.
Expected Results: Biochanin A is expected to attenuate MTX-induced oxidative stress, inflammation, apoptosis, and tissue damage in the kidney and jejunum, with potential dose-dependent effects.
Conclusion: Biochanin A may exert protective effects against MTX-induced nephrotoxicity and jejunal mucositis, potentially through antioxidant, anti-inflammatory, and anti-apoptotic mechanisms.
