Abstract
Background: Spanlastics are surfactant-based nanovesicles that are elastic and deformable, formed from surfactants and edge activators that self-assemble in the water phase. Spanlastics are a new drug-delivery system in which a drug is entrapped in a bilayer within the core cavity. Olanzapine is an atypical antipsychotic drug, which is utilized for the management of schizophrenia, manic depression, and acute mania. Olanzapine has low water solubility and belongs to biopharmaceutical classification class II (low solubility, high permeability). Objective: In this study, olanzapine was engineered to be spanlastic for improved solubility and dissolution rate. Method: Surfactants Span®60 and various edge activators (Soluplus, Cremophor RH, Cremophor EL, Tween 80, and polyvinyl alcohol (PVA)) in different ratios and prepared as spanlastics using the ethanol injection method.
Results: The prepared formulations were tested for particle size, polydispersity index PDI, drug entrapment efficiency, and in vitro release profile. The selected formula underwent additional analysis of zeta potential, field-emission scanning electron microscopy, and compatibility studies using differential scanning calorimetry. The optimized formulation, designated as F4, exhibited a particle size of 93.44±4.7 nm, a polydispersity index of 0.272±0.01, an entrapment efficiency of 83.94±2.03%, and a zeta potential of -33.6±1.8. In vitro release experiments indicated that the optimized formulation (F4), containing PVA as an edge activator, released 100% of the medication over 7 hours. Conclusion: The findings indicate that spanlastics is a promising technique for developing stable and effective drug delivery systems; solubility and dissolution rate increase significantly with spanlastics in comparison with the pure drug
