Abstract

Abstract
Background: Daunorubicin, anthracycline anti-cancer drug, used in acute myeloid leukemia management. Cardiotoxicity is Daunorubicin main systemic toxicity that mediated through increase cardiomyocytes oxidative stress which is Iron mediated. Myocardial damage can be predicted through the rising serum cardiac troponins levels (T and I, subtypes). Turkesterone is a phytoecdysteroid compound that is a 20-hydroxyecdysone insect steroid hormone analogue which has neuroprotective, nephroprotective, anabolic, antidiabetic, and antihyperlipidemic biological effects. Objective: This study explores the protective effects of Turkesterone versus daunorubicin inducing cardiotoxicity in rats. Method: Twenty-four adult Sprague- Dawley rats of both sexes. Animals are divided into four groups of six (6) rats each: Group Ӏ: received (5%) dimethyl sulfoxide for fifteen days. Group ӀӀ: received (12 mg/kg) Daunorubicin last 3 days by IP injection with a total dose of (36 mg/kg). Group III: received Turkesterone (5mg/kg) daily by oral gavage for fifteen days. Group ӀV: received Turkesterone (5mg/kg) daily by oral gavage for fifteen days and then received (12 mg/kg) Daunorubicin last 3 days by IP injection with a total dose of (36 mg/kg). Results and Conclusion: Daunorubicin treatment resulted in clear evidence of myocardial injury, as indicated by the significant increase of serum cardiac troponin I, together with alterations in cardiac GPX4, a key regulator of lipid-peroxide detoxification and ferroptosis-related oxidative injury, inflammation, and apoptosis. Pretreatment with Turkesterone significantly decreased serum cardiac troponin I and decreased daunorubicin-induced changes in cardiac GPx4, interleukin-6 and cleaved caspase-3 levels. The results suggest that Turkesterone may exert a protective effect against daunorubicin-induced cardiac injury, possibly through the maintenance of GPx4-mediated protection against lipid peroxidative damage and the regulation of inflammatory and apoptotic responses.
