Abstract
The aim of this study was to develop and characterize posaconazole-loaded solid lipid nanoparticles (Posa-SLNs) for transdermal drug delivery using a 2³ full factorial design. Solid lipid nanoparticles have a colloidal carrier system made up of solid lipid and surfactant in which the drug is encapsulated in carrier system. Posaconazole is a structural derivative of itraconazole, a related drug. It contains the BCS category of Class II (low solubility/high permeability) and is lipophilic in nature. Posaconazole-loaded solid lipid nanoparticles were synthesized by a high-speed homogenization/ultrasonication method utilizing 23 factorial designs. The standard calibration curve of posaconazole was prepared as well as the equation, and R2value was [y =0.0256x -0.001676; R2=0.9997]. Posaconazole and polymer compatibility studies were studied such as FTIR, DSC, and X-Ray diffraction. The independent variables were glyceryl monostearate, tween 80 and homogenization time. On the basis of responses, these data obtained170 nm particle size, 88% entrapment efficiency and 64% cumulative drug release, Posa-SLNs optimized on it. According to this report, Posa-SLNs are the best carrier system for conveying the posaconazole drug at transdermal delivery for fungal infection.
